Although D40 went into active production last year under the Y Not Studios banner, the project was first announced by Dhanush after Karthik Subbaraj's Iraivi had released in 2016. Production on the film took more than two years to kick-off due to a line of projects both Dhanush and Karthik Subbaraj were already committed to.

The film's plot is set in locations in and around Tamil Nadu and the United Kingdom with excitement shooting up after Dhanush was spotted sporting the Madurai keda moustache as similar to his father-in-law and iconic actor 'Superstar' Rajinikanth in Petta. D40 also includes actress Aishwarya Lekshmi playing the female lead with Sanchana Natarajan, Kalaiayarasan and National Award-winning Malayalam actor Joju George among others in major starring roles.
